Q1. Which is the most common stain is used to stain blood films or smears?

Ans. Leishman’s stain

Q2. What should be the pH of buffer while staining blood smears (using Leishman stain)?

Ans. A pH of 6.8 is recommended for general use.

Q3. Which type of smear we should make for the identification of parasites?

Ans. Thick smear

When parasites are scanty, then thick films are very useful for the identification of parasites.

Q4. What should be the pH of buffer for identification for malaria parasites?

Ans. A pH of 7.2 is recommended for the identification of malaria parasites in order to see schuffner’s dots.

Q5. How to do Leishman’s stain on blood smear?

Ans. First of all dry the blood smear in the air then pours the stain on the slide. It should not stick with slides. After 2 minutes add double the volume of buffer. Mix well and stain for 10 to 15 minutes. Wash it properly with running water.

Q6. What should be the angle of the spreader while making a blood smear?

Ans. 45 degree.

Q7. What is the ideal length and width of a blood smear?

Ans. An ideal smear should be tongue-shaped. The length of the smear should be 3 to 4cm. width of the smear should be 2cm.

Q8. How to prepare Leishman stains?

Answer. Weigh 0.15 gm Leishman stain power per 100 ml of methanol. Mix it stain powder in mortal with the help of pestle in a small amount of methanol. Grind it well & pour the suspension into a big container. Store it in a brown bottle at 37 degrees temperature.

Q9. What is the pH of distilled water?

Ans. 7.0

Q10. How do you report differential leucocyte count?

Ans. We report DLC in percentage.
