What is Improved Neubauer Counting Chamber? | How to use Neubauer Counting Chamber for counting Blood Cells?
The hemocytometer was invented by Louis Charles Malasoez. Neubauer Chamber’s counting grid is 3mm * 3mm in size. The cell counting chamber is divided into 9 squares each of 1mm and an area of 1 square millimeter. The four corners of the counting chamber are further divided into 16 small squares. The central square that is used to see manual platelet count and red blood cells count is divided into 25 medium squares and each medium square with 16 small squares inside.
The depth or height of the chamber is 0.1 mm. when we calculated the volume of one large square then:-
Length of 1 square:- 1mm
Breadth of 1 square – 1mm
Depth of chamber – 0.1 mm
The volume of one large square will be
= l * b * h
= 1mm * 1mm * 0.1mm
= 0.1 per cubic millimeter.
How to count in Neubauer improved cell counting chamber?
First of all, we have to focus on the 10 x objective of the microscope. We can count Total Leucocytes Count or TLC in 10 x objective.
The four corners square in the Neubauer chamber is used for White Cell Count or Total Leucocytes Count. The central square is used for platelets and red cell count.
Procedure to count in Neubauer Chamber
- Place the Neubauer chamber on the microscope & fix with the help of fixing clamp.
- First, focus on 10 x objective with condenser at down position. Try to focus fine lines of Neubauer chamber with fine adjustment.
- For TLC start counting in corner’s four square. First focus one corner square and count cells.
- Write down the number of cells counted in one square then repeat the process for the rest of remaining squares. Add all the cells & multiply with factor that is different for TLC, RBC and platelets counts depending upon dilution factor and the number of squares in which we are counting cells.
The Neubauer chamber is a thick glass slide with a measurement of 30 * 70mm * 4mm (thickness).
The chamber’s central part has double chamber or two counting areas in which we can load two samples simultaneously.
The glass coverslip is required to cover the Neubauer chamber width of 22mm. the glass coverslip is put on the top of the Neubauer chamber, covering the central area.
